Resumen
Heteroatom-tagged proteomics enables the absolute quantification of selenoproteins by using the heteroatom (selenium) as a tag for selective, sensitive atomic detection rather than molecular detection. Using this approach, approximately 90% of total Se in human serum/plasma can be quantified as selenoproteins and total selenometabolites, allowing assessment of Se status. Here, we applied HPLC-ICP-MS to determine absolute concentrations of circulating selenoproteins in colorectal cancer (CRC) patients and healthy controls (HC). An observational study was conducted, including 103 human serum samples from CRC patients (66) and healthy controls (37). Extracellular glutathione peroxidase (eGPx), selenoprotein P (SELENOP), selenoalbumin (SeAlb) and total selenometabolites (SeMT) were quantified, and associations among selenoproteins were evaluated as potential disease biomarkers. eGPx concentrations were significantly higher in CRC patients, whereas SELENOP levels were significantly lower than in HC. Good values of AUC were determined for SELENOP and SeAlb, suggesting their possible utility in clinical studies. Moreover, significant and strong correlations between SELENOP, eGPx, SeMT and SeAlb were observed in HC, but they in CRC patients. Likewise, significant and specific associations were also found between selenoproteins and metals in HC and CRC patients. This study provides the first heteroatom-tagged proteomics–based characterization of selenoproteins in CRC and their comparison with controls, highlighting the potential of the circulating selenoproteome as a source of biomarkers for CRC diagnosis, risk stratification, and therapeutic monitoring.
