Resumen
It is now possible to genetically characterize Peruvian horse breeds such as the Andean Horse, the Morochuco Horse and the Peruvian Paso Horse by genotyping by sequencing (GBS). This methodology requires high-quality DNA. The extraction of horse DNA from blood is an invasive process. The aim of the study was to standardize a method for obtaining high-quality DNA for genomic studies in horses from tail and mane hair. Hair and blood were collected from 10 Creole mares from the Amazon region to evaluate four quantities of hair follicles (T1=80, T2=60, T3=40, T4=20) and 200 µl of blood. Genomic DNA was extracted, and the quantity, quality and integrity of the DNA obtained were analysed by spectrophotometry and agarose gel electrophoresis, respectively. Quality was also evaluated by PCR amplification of the COX1 gene. A normal distribution of the concentration data (ng/µl) was obtained. The best treatment for obtaining DNA was T1, with average concentrations of 22.99 ng/µl and 280/260 and 260/230 ratios of 1.84 and 1.76, respectively. The concentration and purity of the DNA was optimal according to the parameters required by the sequencing platforms. The amount of DNA obtained from blood was higher; however, the hair collection methodology is not invasive, and the transport and storage do not require special conditions unlike other tissues such as blood. © Los autores.
