Resumen
The aim of this study was to evaluate the effect of plasma membrane functionality on cryopreserved boar sperm in samples with different percentages of viable gametes. Fifteen ejaculates were used in four treatments with 100, 75, 50, and 25% viable sperm. Prior to cryopreservation, sperm were incubated for 24 h. Each treatment was diluted in TRIS-Y (Tris with egg yolk), and sperm were stored at 5 °C. TRIS-Y-G-E (Tris with egg yolkglycerol- Equex) was then used for dilution, and sperm were packaged in 0.5 mL straws. Freezing was performed manually in liquid nitrogen in two temperature stages. The straws were thawed at 37 °C for 20 s. Sperm functionality was assessed post-thawing (30 and 150 min) using [Merocyanine 540 (M540) and Yo-Pro-1] in samples incubated in capacitated medium (37 ºC and 5% CO2). Lipid peroxidation was measured spectrophotometrically and indirectly from malonyldialdehyde (MDA) concentrations and intracellular reactive oxygen species (ROS) production in samples incubated for 30 min. MDA concentrations increased as the percentage of viable sperm decreased. A negative correlation was observed between MDA and total motility (TM), progressive motility (PM), and plasma and acrosomal membrane integrity. There was an effect of viable gamete treatment and post-thawing time on ROS production and plasma membrane destabilization. The percentage of viable gametes in the semen before cryopreservation determined the sperm quality of the cryopreserved samples. © (2025), (Universidad Nacional Mayor de San Marcos).
